VHH sequence evidence, strengthened
A review of Selection of Nanobodies Using Sequence Features informed an upgrade to VHH sequence checks and evidence reporting
What changed
- Classical F/Y framework residues are recognized, and CDR3 insertion residues are retained
- Strict checks require native numbering; approximate annotations remain explicitly uncertain
- 11 source-linked knowledge records distinguish reported observations from validation proposals
- A sequence-check panel reports IMGT42, IMGT123 and CDR3 length before design
How to interpret it
Sequence features do not prove binding, stability or pH response. The patent's overall F/Y ELISA comparison was not significant; the favorable result was in a two-cysteine subgroup. Its MSLN example does not establish CAR-T efficacy
New feature annotations do not change ranking weights. Corrected validation can change which candidates pass. VHH Design remains restricted while broader execution validation continues
